AB AIM: To characterise the induction of proinflammatory cytokines by C.
neoformans and to determine if complement is an essential factor in this
induction. METHODS: Peripheral blood mononuclear cells (PBM) were
obtained from normal hosts and stimulated in vitro with either killed,
whole C. neoformans or lipopolysaccharide. All steps in the in vitro
assays were performed under conditions to minimise endotoxin
contamination. RESULTS: Experiments showed that peak levels of IL-1
beta, B, TNF-alpha and IL-6 were detected when first measured at 10 hrs
of stimulation with C. neoformans. IL-1 beta and TNF-alpha levels
declined after 24 hrs whereas IL-6 production persisted up to 9 days.
The kinetics and magnitude of responses to C. neoformans were similar to
LPS. Experiments showed that lower concentrations of cryptococcus had
similar TNF-alpha production with both heat inactivated and normal sera,
but was decreased in the absence of sera. However, at higher
concentrations the TNF-alpha production was independent of the presence
of sera. CONCLUSIONS: C. neoformans induces the production of
proinflammatory cytokines with similar kinetics to LPS. The induction of
TNF-alpha was independent of complement.
